Abstract
U rasadničkoj proizvodnji važnu ulogu ima kontinuirano uvođenje suvremenih in vitro metoda klonskog razmnožavanja reprodukcijskog materijala. Poncirus trifoliata Raf. je najzastupljenija i najvažnija podloga mandarine Unšiu u Hrvatskoj i drugim proizvodnim zemljama svijeta. Istraživanje o mogućnosti mikropropagacije vegetativne podloge agruma Poncirus trifoliata Raf. provedeno je na Fakultetu agrobiotehničkih znanosti Osijek tijekom 2020. godine. Ispitivan je utjecaj pojedinih biljnih regulatora rasta (BAP, IBA, Zeatin i GA3) na mogućnost proliferacije i organogeneze nodijalnih eksplantata iniciranih na MS hranjivi medij. Najbolji rezultati dobiveni su na tretmanu T1 koji je uključivao kombinaciju citokinina BAP i auksina IBA. Na ovom tretmanu zabilježena je najmanja stopa kontaminacije i odumrlih eksplantata. Zdravi eksplantati T1 tretmana uključeni su u daljnju fazu multiplikacije. Nakon nekoliko uspješnih ciklusa multiplikacije dobiven je dovoljan broj novih zdravih mikrobiljaka na kojim će se obavljati brojna druga istraživanja.
Abstract (english)
The continuous introduction of modern in vitro methods in clonal propagation of plating material plays an important role in nursery plants production. Poncirus trifoliata Raf. is the most represented and most important rootstock of mandarin Unshiu in Croatia and other production countries. Research on the possibility of micropropagation citrus vegetative rootstock Poncirus trifoliata Raf. was conducted at the Faculty of Agrobiotechnical Sciences Osijek during 2020. The influence of individual plant growth regulators (BAP, IBA, Zeatin and GA3) on the possibility of proliferation and organogenesis of nodal explants initiated on MS nutrient medium was investigated. The best results was obtained on T1 treatment, which included a combination of cytokine BAP and auxin IBA. This treatment has the lowest rate of contamination and dead explants. Healthy explants of T1 treatment are included in the further phase of multiplication. After several successful multiplication cycles, a sufficient number of new healthy microplants have been successfully obtained on which numerous other researches will be performed.
