Abstract
Polifenol oksidaza (PPO) je enzim koji utječe na kvalitetu pšenice i proizvoda od pšenice. Aktivnost polifenol oksidaze kontrolirana je PPO genima, od kojih su najviše proučavani oni smješteni na kromosomima pšenice 2AL i 2DL. Primjena molekularnih markera, koji su povezani s glavnim PPO genima, ima važnu ulogu u selekciji genotipova s niskom aktivnosti PPO. Cilj ovoga istraživanja bio je utvrditi zastupljenost Ppo-A1 i Ppo-D1 gena u hrvatskom sortimentu pšenice. Istraživanje je provedeno na 30 hrvatskih sorti pšenice. PCR analiza provedena je korištenjem tri para početnica za analizu Ppo-A1 i Ppo-D1 lokusa. Produkti amplifikacije analizirani su horizontalnom elektroforezom. Na Ppo-A1 lokusu, Ppo-A1a alel utvrđen je kod 43,33 % ispitivanih sorti pšenice, a alel Ppo-A1b kod njih 56,67 %. Na Ppo-D1 lokusu, Ppo-D1a alel utvrđen je kod 63,33 % ispitivanih sorti pšenice, a alel Ppo-D1b kod njih 36,67 %. Najveću učestalost imala je kombinacija alela Ppo-A1b i Ppo-D1a koja je zabilježena kod 40 % ispitivanih sorti pšenice.
Abstract (english)
Polyphenol oxidase (PPO) is an enzyme that affects the quality of wheat and wheat products. Polyphenol oxidase activity is controlled by PPO genes, with the most studied being those located on wheat chromosomes 2AL and 2DL. The use of molecular markers, which are related to the main PPO genes, has an important role in the selection of genotypes with low PPO activity. The aim of this research was to determine the presence of Ppo-A1 and Ppo-D1 genes in the Croatian wheat varieties. The research was conducted on 30 Croatian wheat varieties. PCR analysis was performed using three pairs of primers for the analysis of the Ppo-A1 and Ppo-D1 loci. Amplification products were analyzed by horizontal electrophoresis. At the Ppo-A1 locus, the Ppo-A1a allele was present in 43.33% of the examined wheat varieties, and the Ppo-A1b allele in 56.67% of them. At the Ppo-D1 locus, the Ppo-D1a allele was present in 63.33% of the tested wheat varieties, and the Ppo-D1b allele in 36.67% of them. Combination of alleles Ppo-A1b and Ppo-D1a had the highest frequency and was recorded in 40% of the tested wheat varieties.
