Abstract
Proizvodnja borovnice (Vaccinium corymbossum L.) u RH je zanemariva iz razloga što je relativno nova intenzivna kultura kojoj nedostaje dovoljno iskustva od strane domaćih proizvođača. Mikropropagacija, odnosno tehnika kulture tkiva in vitro osigurava dobivanje visokokvalitetnog i fitosanitarno ispravnog sadnog materijala u kratkom vremenskom intervalu, a proizvodnja se odvija u kontroliranim i aseptičnim uvjetima. Pokus je postavljen u in vitro labaratoriju za voćarstvo na Fakultetu agrobiotehničkih znanosti Osijek tijekom 2020. godine. Cilj ovog završnog rada usmjeren je na ispitivanje mogućnosti organogeneze eksplantata borovnice u kulturi tkiva in vitro. Tretmani su uključivali dva tipa eksplantata: bazni i vršni dio izdanka. Ispitivan je i utjecaj broja eksplantata po teglici (25 i 50) na organogenezu i multiplikaciju budućih mikroizdanaka. Rezultati ukazuju kako veći broj eksplantata unutar in vitro posude nema štetan utjecaj na daljnju fazu multiplikacije (multiplikacija je povećana). Vršni tip eksplantata rezultirao je većim brojem izdanaka i listova, dok je visina izdanaka i multiplikacija bila veća na baznom tipu eksplantata. Prema dobivenim rezultatima zaključujemo kako nema velike razlike u organogenezi između pojedinih tipova eksplantata borovnice (vršni ili bazni dio). Nastale mikroizdanke s oba tipa eksplantata moguće je uključiti u daljnju fazu multiplikacije i/ili rizogeneze.
Abstract (english)
The production of blueberries (Vaccinium corymbossum L.) in Croatia is negligible due to the fact that it is a relatively new intensive fruit crop with lack experience of domestic producers. Micropropagation ensures the production of high quality and phytosanitary safe planting material in a short time interval with production in controlled and aseptic conditions. The whole experiment was carried out on in vitro laboratory for fruit growing, Faculty of Agrobiotechnical Science Osijek due to the 2020 year. The aim of this BSc Thesis is to examine the possibility of organogenesis blueberries explants in tissue culture in vitro. Treatments included two types of explants: the base and the apical part of the shoot. The influence of the number of explants per in vitro jar (25 and 50) on organogenesis and multiplication rate of future microshoots was also investigated. The results indicate that a greater number of explants inside the in vitro jar (50) has no harmful influence in the further phase of multiplication (multiplication is increased). Apical type of explant resulted in more shoots and leaves, while shoot height and multiplication rate was greater on the base type of explants. According to the obtained results, we conclude that there is no greater difference on the organogenesis between explants types. New microshoots from both type of explants can be included in the further phase of multiplication and/or rhizogenesis.
