Abstract
Cilj ovog istraživanja bio je ispitati alelnu varijabilnost Ppd-D1 lokusa za reakciju na duljinu dana u 20 hrvatskih ozimih pšenica pomoću specifičnih mikrosatelitnih početnica, te rezultate usporediti s datumom klasanja u poljskim pokusima. Identificirane su dvije različite alelne varijante na Ppd-D1 lokusu: Ppd-D1a od 288bp i Ppd-D1b od 414 bp. Alelna varijanta Ppd-D1a bio je prisutan u 18 od 20 (90%) sorata uključenih u istraživanje, što je u skladu sa dosadašnjim istraživanjima. Alelna varijanta Ppd-D1b detektirana je u samo jednoj sorti, sorti Dea. Obje alelna varijante, Ppd-D1a i Ppd-D1b, pronađene su u sorti Barbara što ukazuje na heterozigotnost ove sorte na ispitivano svojstvo. Rezultati dobiveni uspoređivanjem datuma klasanja i prisutnosti Ppd-D1a, odnosno Ppd-D1b alela su nedosljedni, što je najvjerojatnije rezultat ekstremnih klimatskih uvjeta tijekom vegetacijske godine 2013./2014..
Abstract (english)
The aim of this study was to determine the variability of Ppd-D1 locus for photoperiod response in 20 Croatian winter wheat varieties using specific microsatellite primers and to compare the results with heading date wheat heading date obtained from field trial. Two different alleles were determined on Ppd-D1 locus Ppd-D1a from 288bp and Ppd-D1b from 414 bp. Among 20 cultivars, 18 (90%) carried photoperiod-insensitive allele Ppd-D1a. One cultivar, Dea, carried photoperiod-sensitive Ppd-D1b allele. Both photoperiod-insensitive and photoperiod-sensitive alleles were found in cultivar Barbara, what suggests that this cultivar is heterozygous for this trait. When comparing heading date and Ppd-D1a and Ppd-D1b allele distribution, results of this study were inconclusive, most likely because extreme climate conditions during vegetation season 2013/2014.
