Abstract
Cilj istraţivanja bio je ispitati i utvrditi utjecaj različitih metoda sterilizacije sjemenki zbog odreĎivanja najučinkovitijih postupaka za dobivanje sadnica Calendula officinalis in vitro metodom klijanja sjemenki na hranjivoj podlozi. U Istraţivanju je ispitano djelovanje etanola i varikine u različitim koncentracijama i vremenskim intervalima na dezinfekciju sjemenki nevena. Za pokus korišteno je komercijalno sjeme Calendula officinalis iz Njemačke kupljeno u trgovačkom centru. Pokus se sastojao od 8 tretmana i kontrolnog tretmana. Svaki tretman bio je zastupljen s 5 Erlenmeyer tikvica s hranjivom podlogom na koju se uvodilo po 5 sjemenki. Utjecaj različitih tretmana nije se ispostavio učinkovitim jer se kontaminiralo 44 od 45 hranjivih podloga. Najveća zabiljeţena kontaminacija bila je na kontrolnom tretmanu (sjemenke koje su bile samo u vodi 30 minuta), gdje su sve hranjive podloge bile kontaminirane i s bakterijom i gljivicom. Hranjiva podloga koja se nije kontaminirala sadrţavala je sjemenke koje su bile drţane 30 minuta u otopini varikine u koncentraciji od 10 %.
Abstract (english)
The goal of the research was to examine and determine the impact of different methods of seed sterilization in order to determine the most effective procedures for obtaining Calendula officinalis seedlings in vitro by the method of seed germination on a nutrient medium. In the research, the effect of ethanol and bleach in different concentrations and time intervals on the disinfection of marigold seeds was examined. Commercial Calendula officinalis seeds from Germany bought in a shopping center were used for the experiment. The experiment consisted of 8 treatments and a control treatment. Each treatment was represented by 5 Erlenmeyer flasks with a nutrient medium to which 5 seeds were introduced. The influence of different treatments did not turn out to be effective because 44 out of 45 nutrient media were contaminated. The highest recorded contamination was in the control treatment (seeds that were only in water for 30 minutes), where all nutrient media were contaminated with both bacteria and fungus. The nutrient medium that did not become contaminated contained seeds that were kept for 30 minutes in a 10 % bleach solution.
