Abstract
Pšenica je iza kukuruza druga najznačajnija žitarica iz porodice trava. Cilj ovog rada bio je uspješno izolirati genomsku DNA iz mljevenog zrna pšenice bez primjesa. Samo istraživanje provelo se u dvije faze: u prvoj fazi provedene su dvije metode izolacije DNA iz zrna pšenice, a to su modificirana CTAB metoda s dodatkom PVP i SDS metoda, a DNeasy® Plant Mini Kit test korišten je kao brza provjera kvalitete DNA . Druga faza sastoji se od spektrofotometrijske analize izolirane DNA. Modificirana CTAB metoda se pokazala kao najuspješnija s optimalnom čistoćom i koncentracijom izolirane DNA (A260/280 2,00 – 1,99, A260/230 2,19 – 2,34, koncentracija 97,10 – 181 ng/μL).
Abstract (english)
The wheat besides corn is the world the second most important crop of the grass family. The aim of this work was to successfully isolate genomic DNA from milled wheat grains without secondary methabolutes. The research was carried out in two phases: in the first phase were used two DNA isolation methods: modified CTAB method with PVP supplement and SDS method, and the DNeasy® Plant Mini Kit test was used as a quick DNA quality check. Second phase consist of spectrophotometric determination isolated DNA. Modified CTAB method was most successful generating optimal DNA quality and quantity results (A260/280 2.00 – 1.99, A260/230 2.19 – 2.34, concentration 97.10 – 181 ng/μL).
