Abstract
Restrikcijska endonukleaza je naziv za enzim pronađen u bakterijskim stanicama kao dio njihova restrikcijsko modifikacijskog sustava koji im služi u obrani i razgradnji virusne DNA. Cilj ovog istraživanja je bio izolirati genomsku DNA iz tri biljne vrste s različitom veličinom genoma te provesti uspješnu digestiju genomske DNA restrikcijskim endonukleazama. U radu su korištene vrste Brachypodium distachyon L. i kukruz (Zea mays L.) i šest restrikcijskih endonukleaza. Genomska DNA izolirana je koristeći CTAB metodu, a digestija izolirane DNA je optimizirana te su rezultati cijepanja provjeren elektroforezom. Utvrđeno je da su najpogodniji enzimi za cijepanje genosmke DNA Brachypodium distachyon L., EcoRI i BamHI, a za Zea mays L. to su enzimi HindIII, RspRSII, EcoRI, pri uvjetima inkubacije i volumenu sastavnica restrikcijskih smjesa propisanih od strane proizvođača.
Abstract (english)
Restriction endonuclease is the name for an enzyme found in bacterial cells as part of their restriction modification system that are used in defense and degradation of foreign DNA. The aim of the study was to isolate genomic DNA from three plant species with different genome sizes and to perform successful digestion of genomic DNA with restriction endonucleases. The species Brachypodium distachyon L. and maize (Zea mays L.) and six restriction endonucleases were used in the work. Genomic DNA was isolated using the CTAB method, digestion of the isolated DNA was optimized and the cleavage results were verified by electrophoresis. It was determinedthat the most suitable enzymes for genomic DNA of Brachypodium distachyon L. are EcoRI and BamHI, and for Zea mays L. to su enzimi HindIII, RspRSII, EcoRI, under the conditions of incubation and the volume of components of the restriction mixtures prescribed by the manufacturer.
